Antibody and Protein Labeling

Antibody labeling is an important technique to aid in the detection or isolation and purification of a protein. Labeled antibodies are essential tools in applications of biological research and diagnostics, such as western blots, enzyme-linked immunosorbent assay (ELISA), immunofluorescence (IF), and immunohistochemistry (IHC).
Protein labeling refers to the use of the appropriate molecular labels to facilitate the detection or purification of a labeled protein and its binding partners.

Reporter enzymes (such as HRP) can be attached to antibodies or proteins using different coupling chemistries to ensure the maximum retention of activity of both the enzyme and the protein. Non-enzymatic biotin, isotopes, and FITC can be coupled to specific antibodies, proteins, and peptides as a means of signal generation.

Biotin

Biotin is a small molecule (244.3 Da) with a wide distribution in animal and plant tissues. As the affinity of avidin or streptavidin for biotin is one the strongest known non-covalent interactions, biotin-labeled antibodies can increase the sensitivity of an assay and be employed in detection (such as ELISA, western blot, IF, and IHC).

With the development of mass spectrometer and computers, the stable isotope labeling technique provides a vision for the understanding of absolute information about all proteins in a biological sample. There are two popular approaches; one is the absolute quantification method (AQUA) using stable isotope-labeled peptides as internal standards, and the other is metabolic stable isotope labeling in amino acids in cell culture (SILAC).

HRP

HRP-labeled antibodies can react with specific substrates, generating a chromogenic, chemiluminescent, or a fluorescent signal upon the oxidation ability of HRP, which makes HRP to be one of the most commonly used enzyme labels in the area of antibody-based antigen detection assays.

FITC

Fluorescent dyes have a wide range of selections, which provides greater flexibility, variation, and performance for research applications. Fluorescein, based on its amine-reactive isothiocyanate derivative FITC, has been one of the most popular fluorophores because of its high absorptivity, excellent fluorescence quantum yield, and good water solubility. Unlike biotin or HRP, FITC molecule could be utilized alone and does not require additional reagents for detection, which makes FITC extremely versatile for monitoring biological processes in vivo.
