scFv/Fab

The construction of scFv is usually the first step of antibody engineering. scFv antibody fragments have several advantages compared with monoclonal antibodies such as low kidney uptake, rapid blood clearance, greater tumor penetration, and less negative response from the human immune system. Fab fragments are one of the most common constructions of antibody engineering. These antibody fragments differ from scFv's as they are composed of one constant and one variable domain of each of the heavy and the light chains.

Chimeric Antibody

As one of the most important antibody engineering methods, antibody chimerization has been widely used in therapeutic and diagnostics applications or researches. A chimeric antibody is an antibody made by combining genetic material from one species source, like a mouse, with genetic material from another species, such as a human being. Chimeric antibodies retain the antigen specificity and affinity of the foreign antibody, and are less easily rejected by the body's immune system compared with the ordinary monoclonal antibody.

Antibody Humanization

Antibody humanization could decrease the immunogenicity of monoclonal antibodies and maintain high specificity. Unlike the chimeric antibody, the protein sequence of a humanized antibody is essentially identical to that of a human variant, despite some of its complementarity determining region (CDR) segments being of non-human origins. With the development of targeted anticancer drugs, humanized antibodies are essential in drug development and therapeutic application.

Fc Fusion Proteins

Fc fusion proteins are composed of an immunoglobulin Fc domain that is directly linked to another peptide or protein of interest. Fc fusion proteins can play a role in dimerization, in vivo half-life, and receptor activation and affinity for the cognate ligand.

Antibody library is based on the technique of phage display. Compared with the traditional hybridoma method, phage display library has enormous advantages in discovering novel antibodies for therapeutic and diagnostics applications or studies. We have non-immune libraries (naive and synthetic libraries, containing 1 billion to 1 trillion independent clones) and immune libraries (at least 5-100 billion independent clones).

Antibody Sequencing

An antibody with a known protein sequence could be used for recombinant expression, patent application, downstream development, etc. Hybridoma sequencing, also known as antibody sequencing, is the sequencing of the cDNA encoding the variable heavy (VH) and variable light (VL) domains of your hybridoma cell line. BIC offers a professional and a highly versatile antibody sequencing service, and the sequencing range covers mice, rats, rabbits, goats, chickens, and other species.

Affinity Maturation

After humanization, the affinity of antibodies may drop significantly. In some other cases, an antibody needs to reach a minimum affinity level, or the final application requires extremely high antigen-binding affinity. In these cases, enhancing the affinity of the interest antibody will be the best approach that could take the antibody into clinical development. In addition, it can be useful to remove potential modification sites for improved developability.
