TEV protease (Tobacco Etch Virus Protease), Protease Recombinant, E.coli

The tobacco etch virus (TEV) protease is an important enzyme for life science research. The widely used TEV protease is the 27kDa catalytic domain of the nuclear inclusion an (NIa) protease from tobacco etch virus. Its high specificity and robustness make it ideal for diverse applications. It is used in vitro for protein purification and in vivo to test for protein–protein interactions, for induced proteolysis and to generate conditional mutants. Among various proteases, TEV protease outstands because of its high and unique specificity. It can be used at temperature as low as 4°C with adequate efficiency to reduce the proteolysis of the target protein. The biological function of the protease is to proteolyse the viral polyprotein into single proteins during tobacco etch virus biogenesis. Nowadays, it is used more frequently than other proteases (enterokinase, thrombin, factor Xa, and human rhinovirus 3C protease) in structural genomic research projects.
Description Recombinant TEV Protease (rTEV) is a site-specific protease purified from E. coli. The protease can be used for the removal of affinity tags from fusion proteins. The seven-amino-acid recognition site for rTEV is Glu-Asn-Leu-Tyr-Phe-Gln-Gly with cleavage occurring between Gln and Gly. The optimal temperature for cleavage is 30°C; however, the enzyme can be used at temperatures as low as 4°C.
The rTEV contains His tag.
The rTEV is purified by proprietary chromatographic techniques.

The rTEV contains 0.50M Tris-HCl pH 8.0, 10mM DTT and 5mM EDTA.
Stability The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles.
Unit Definition One unit of rTEV cleaves >85% of 3 ug control substrate in 1 h at 30°C.
