IL-11 (Interleukin-11), Human Recombinant, Pichia Pastoris

Interleukin-11 (IL-11) is a pleiotropic cytokine that exhibits a wide variety of biological activities which are determined by expression of IL-11Rα and gp130 on the cell membrane in neutrophils as well as in the haematopoietic and immune systems. Interleukin 11 (IL11) belongs to the IL6-type family of cytokines and signals via a heterodimeric receptor complex comprising the specific IL11 receptor alpha (IL11RA) chain and the associated signaling subunit. IL-11 influences both cellular proliferation and differentiation and also importantly has roles in tumorigenesis, stimulating migration or invasion of gastric, colorectal, and breast carcinomas. Interleukin 11 also promotes formation of active GTP-bound Ras and induces activation of mitogen-activated protein kinases (MAPKs), with cross-talk between Ras-MAPK and JAK/STAT signaling pathways also occurring.
Description IL11 Human Recombinant produced in Pichia Pastoris is a single, non-glycosylated, Polypeptide chain containing 177 amino acids (it differs from the 178 amino acid length of the native IL11 only in lack of the N-terminal praline residue) and having a molecular mass of 19kDa.
The IL11 is purified by proprietary chromatographic techniques.

Amino Acid Sequence The sequence of the first five N-terminal amino acids was determined and was found to be Gly-Pro-Pro-Pro-Gly.
Purity >96% as determined by SDS-PAGE and HPLC
Formulation Sterile Filtered White lyophilized (freeze-dried) powder.
IL11 was lyophilized after extensive dialysis against 20mM PB, pH7.0 and 2% Glycine buffer.
Stability The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles.
Biological Activity The ED50 as determined by the dose-dependent stimulation of the proliferation of murine 7TD1 was found to be less then 0.2ng/ml, corresponding to a Specific Activity of 8,000,000 IU/ mg.
Solubility It is recommended to reconstitute the lyophilized Interleukin -11 in sterile 18M?-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
