IL-10 (Interleukin-10), Human Recombinant, HEK

Interleukin 10 (IL-10) is a pleiotropic cytokine, and often essential, moreover, IL-10 is a major immunosuppressive and anti-inflammatory factor that is indispensable for the homeostatic control of infection and inflammation. Key functions include deactivation of myeloid lineage cells and suppression of inflammatory cytokine production. A hallmark action of IL-10 is suppression of cell activation and gene induction by inflammatory factors such as Toll-like receptor (TLR) ligands, and it has been proposed that IL-10 can inhibit TLR signalling. Within the central nervous system, IL-10 is neuroprotective, as evidenced by enhanced in vitrocell culture survival of embryonic or immature cortical neurons, retinal ganglion cells, cerebellar granule cells, spinal cord neurons, or in hippocampal slice preparations from juvenile rats following excitotoxicity.
Description Interleukin-10 Human Recombinant produced in HEK cells is a glycosylated non-disulfide bonded homodimer, having a total molecular weight of 17kDa.
The IL10 is purified by proprietary chromatographic techniques.

Formulation Sterile Filtered White lyophilized (freeze-dried) powder.
The IL10 was lyophilized from 1mg/ml in 10mM Tris-HCl, pH 7.4 and 150mM NaCl.
Stability The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles.
Biological Activity The specific activity was determined by the dose-dependent stimulation of the proliferation of MC/9 cells (mouse mast cell line) in the presence of 200 pg/ml IL-4, and is typically 0.3-1.5ng/ml.
Solubility It is recommended to reconstitute the lyophilized IL-10 in sterile water not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
