IL-2 | Interleukin-2, human

Interleukin (IL)-2, initially described as a T cell growth factor, was an early candidate for the immunotherapy of cancer and approved for use in renal cell cancer by the FDA in 1992. IL-2 controls the amplification of T cells by initially stimulating growth following antigen activation, and then by promoting the activation-induced cell death. The ability of IL-2 to stimulate NK cell and CD8+ T cell lysis of tumor targets generated early clinical interests for IL-2 as a biological agent for cancer treatment.
Full Name Interleukin-2 Human Recombinant
Synonyms T-cell growth factor (TCGF), Aldesleukin, Lymphokine, IL-2.
Source E. coli
Amino Acid Sequence The sequence of the N-terminal amino acids was determined to be Met-Ala-Pro-Thr-Ser.
Molecular Weight Interleukin-2 Human Recombinant produced was produced in E. coli as a single, non-glycosylated mutein (a variant form with a Ser at position 126 instead of Cys) of human IL-2 polypeptide chain containing 134 amino acids and shown to exhibit a molecular mass of 15 kDa.
Purity >95% as determined by SDS-PAGE and HPLC
Endotoxin Endotoxin level was found to be ≤ 0.1 ng/µg(1EU/µg) by a LAL gel clot method.
Formulation Lyophilized from a sodium monobasic (0.17 mg) & dibasic sodium (0.89 mg) phosphate buffer solution pH7.5, filtered with a membrane (0.2 µm).
Reconstitution To reconstitute the protein, add sterile water to the lyophilized protein and make a preparation with a concentration ≥0.1mg/ml. The preparation can be diluted into other appropriate buffers if necessary. Cautions should be taken to avoid bacterial growth, resulting in potential contamination with endotoxin.
Stability Once lyophilized, the protein is stable at room temperature (≈3 weeks) and at -20°C (≥ 2 years from the date of receipt).Upon reconstitution, it can be stored at 4°C (≤ 7 days) or at -20°C (≤ 6 months). For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended.Avoid frequent freeze-thaw cycles.
Biological Activity The ED50, as determined by a dose-dependent stimulation of murine CTLL-2 cells, is < 0.0645 ng/ml, corresponding to a Specific Activity of 15,500,000IU/mg.
Usage FOR LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes.
