G-CSF (Granulocyte Colony Stimulating Factor), Human Recombinant, E. coli
Granulocyte colony-stimulating factor (G-CSF) is a glycoprotein that induced differentiation of a murine myelomonocytic leukemic cell line. It is widely known as a hematopoietic cytokine that promotes survival, proliferation and differentiation of cells of the neutrophil lineage. G-CSF has been investigated and found able to promote granulocyte precursors and neutrophil differentiation, proliferation, and maturation in the blood. G-CSF able to stimulate both myelopoiesis and hematopoietic stem cell mobilization, which has seen it used clinically for patients with leukocytopenia and for donors of peripheral blood-derived hematopoietic progenitor cells prior to collection for transplantation.
Full Name | Granulocyte Colony Stimulating Factor Human Recombinant |
Synonyms | G-CSF, CSF-3, MGI-1G, GM-CSF beta, Pluripoietin, Filgrastim, Lenograstim, MGC45931, GCSF. |
Source | E. coli |
Amino Acid Sequence | The sequence of the N-terminal amino acids of GCSF was determined to be Met-Thr-Pro-Leu-Gly. |
Molecular Weight | Granulocyte Colony Stimulating Factor Human Recombinant was produced in E. coli as a single, non-glycosylated, polypeptide chain containing 175 amino acids and shown to have a molecular mass of 18.8 kDa. |
Purity | >95% as determined by SDS-PAGE and HPLC |
Endotoxin | Endotoxin level was found to be ≤ 0.1 ng/µg(1EU/µg) by a LAL gel clot method. |
Formulation | Lyophilized from a 0.2 µm filtered 10mM sodium acetate buffer pH 4. |
Reconstitution | To reconstitute the protein, add sterile water to the lyophilized protein and make a preparation with a concentration ≥0.1mg/ml. The preparation can be diluted into other appropriate buffers if necessary. Cautions should be taken to avoid bacterial growth, resulting in potential contamination with endotoxin. |
Stability | Once lyophilized, the protein is stable at room temperature (≈3 weeks) and at -20°C (≥ 2 years from the date of receipt).Upon reconstitution, it can be stored at 4°C (≤ 7 days) or at -20°C (≤ 6 months). For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended.Avoid frequent freeze-thaw cycles. |
Biological Activity | The ED50, determined by the dose-dependent stimulation of proliferation of murine NFS-60 indicator cells (measured by 3H-thymidine uptake), is < 0.1 ng/ml, corresponding to a Specific Activity of 100,000,000 IU/mg. |
Usage | FOR LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes. |
