G-CSF (Granulocyte Colony Stimulating Factor), Human Recombinant, E. coli

Granulocyte colony-stimulating factor (G-CSF) is a glycoprotein that induced differentiation of a murine myelomonocytic leukemic cell line. It is widely known as a hematopoietic cytokine that promotes survival, proliferation and differentiation of cells of the neutrophil lineage. G-CSF has been investigated and found able to promote granulocyte precursors and neutrophil differentiation, proliferation, and maturation in the blood. G-CSF able to stimulate both myelopoiesis and hematopoietic stem cell mobilization, which has seen it used clinically for patients with leukocytopenia and for donors of peripheral blood-derived hematopoietic progenitor cells prior to collection for transplantation.
Full Name Granulocyte Colony Stimulating Factor Human Recombinant
Synonyms G-CSF, CSF-3, MGI-1G, GM-CSF beta, Pluripoietin, Filgrastim, Lenograstim, MGC45931, GCSF.
Source E. coli
Amino Acid Sequence The sequence of the N-terminal amino acids of GCSF was determined to be Met-Thr-Pro-Leu-Gly.
Molecular Weight Granulocyte Colony Stimulating Factor Human Recombinant was produced in E. coli as a single, non-glycosylated, polypeptide chain containing 175 amino acids and shown to have a molecular mass of 18.8 kDa.
Purity >95% as determined by SDS-PAGE and HPLC
Endotoxin Endotoxin level was found to be ≤ 0.1 ng/µg(1EU/µg) by a LAL gel clot method.
Formulation Lyophilized from a 0.2 µm filtered 10mM sodium acetate buffer pH 4.
Reconstitution To reconstitute the protein, add sterile water to the lyophilized protein and make a preparation with a concentration ≥0.1mg/ml. The preparation can be diluted into other appropriate buffers if necessary. Cautions should be taken to avoid bacterial growth, resulting in potential contamination with endotoxin.
Stability Once lyophilized, the protein is stable at room temperature (≈3 weeks) and at -20°C (≥ 2 years from the date of receipt).Upon reconstitution, it can be stored at 4°C (≤ 7 days) or at -20°C (≤ 6 months). For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended.Avoid frequent freeze-thaw cycles.
Biological Activity The ED50, determined by the dose-dependent stimulation of proliferation of murine NFS-60 indicator cells (measured by 3H-thymidine uptake), is < 0.1 ng/ml, corresponding to a Specific Activity of 100,000,000 IU/mg.
Usage FOR LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes.
