Exendin-4 Recombinant, E.coli

Exenatide is an incretin mimetic, a synthetic form of the naturally occurring exendin-4 isolated from the saliva of the Gila monster (Heloderma suspectum). Exenatide has 53% amino acid sequence identity with human glucagon-like peptide-1 (GLP-1), and has been widely used as an adjunctive therapy. Exendin-4 is a molecule currently used, is a glucagon like peptide-1 receptor agonist. Exendin-4 exerts insulinotropic effects and has multiple glucose regulatory functions through activation of GLP-1 receptor in the mammalian cells. It binds and activates the Glucagon-Like Peptide-1 Receptor (GLP-1R), thus stimulation of insulin secretion, suppression of glucagon secretion, enhancement of beta-cell mass, slowing of gastric emptying, and inhibition of food intake.
Description Exendin-4 Recombinant is a novel 39-amino acids peptide having a molecular mass of 4186.7 Dalton, which shares 53%, sequence homology with GLP-17-36 amide and interacts with the same membrane receptor.
The Exendin-4 is purified by proprietary chromatographic techniques.
Synonyms Exenatide.
Source Escherichia Coli.
Amino Acid Sequence The sequence determined and was found to be His-Gly-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Leu-Ser-Lys-Gln-Met-Glu-Glu-Glu-Ala-Val-Arg-Leu-Phe-Ile-Glu-Trp-Leu-Lys-Asn-Gly-Gly-Pro-Ser-Ser-Gly-Ala-Pro-Pro-Pro-Ser.
Purity >96% as determined by SDS-PAGE and HPLC
Formulation Sterile Filtered White lyophilized (freeze-dried) powder.
The Exendin-4 peptide was lyophilized after extensive dialyses against 20mM PBS and contains 4% mannitol.
Stability The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles.
Biological Activity 1. Regulates Glucose levels rapidly
2. Reduces Insulin resistence
3. Reduces Glucagon
4. Reduces HbA1c
5. Stimulates beta cell growth which stimulates insulin production.
Solubility It is recommended to reconstitute the lyophilized Exendin Recombinant in sterile 18MΩ-cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
Usage LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes.
