PTH [Parathyroid hormone (1-34)], Human Recombinant, E.coli
Parathyroid hormone (PTH) is an essential hormone for the regulation of serum calcium levels. PTH is secreted by the parathyroid glands as a polypeptide containing 84 amino acids in response to low serum calcium levels. PTH is one of the major systemic regulators of bone metabolism. It is secreted from the parathyroid glands and travels through the bloodstream to act upon bone. Hypoparathyroidism due to inadequate production or secretion of parathyroid hormone (PTH) leads to hypocalcemia, hyperphosphatemia and, in some cases, hypercalciuria. At the level of the bone, PTH deficiency results in decreased bone turnover and, oftentimes, increased bone mass as measured by DXA and histomorphometry. Several studies demonstrating that the hypocalcemia of hypoparathyroidism can be successfully managed using synthetic human PTH 1-34 (hPTH 1-34) in lieu of vitamin D analogues.
Description | Parathyroid Hormone Human Recombinant (C181H290N55O51S2) produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 34 amino acids and having a molecular mass of 4117.8 Dalton. The PTH is purified by proprietary chromatographic techniques. |
Synonyms | Parathyrin, PTH, Parathormone. |
Source | Escherichia Coli. |
Amino Acid Sequence | The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Val-Ser-Glu-Ile. The amino acid composition is as follows: Ser-Val-Ser-Glu-Ile-Gln-Leu-Met-His-Asn-Leu-Gly-Lys-His-Leu-Asn-Ser-Met-Glu-Arg-Val-Glu-Trp-Leu-Arg-Lys-Lys-Leu-Gln-Asp-Val-His-Asn-Phe. |
Purity | >96% as determined by SDS-PAGE and HPLC |
Formulation | Sterile Filtered White lyophilized (freeze-dried) powder. The protein (1 mg/ml) was lyophilized after extensive dialyses against 1.15 mg sodium citrate, sodium chloride 7.31 mg, 0.21 mg citric acid, 0.1117 EDTA-Na2, 0.2 mg Tween 80 and 50 mg Mannitol. |
Stability | The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles. |
Biological Activity | The activity calculated by UMR106 cell/cAMP method corresponding to a specific activity of 10,000 Units/mg. |
Solubility | It is recommended to reconstitute the lyophilized Parathormone in sterile 18MΩ-cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions. |
Usage | LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes. |
