IL-3 (Interleukin-3), Human Recombinant, E.coli
Interleukin-3 (IL-3) is a pleiotropic cytokine produced principally by activated T-cells that is widely expressed in vivo, and promotes the proliferation, survival and differentiation of multiple hematopoietic cells, such as activated T-lymphocytes, mast cells and basophils. During immune responses that is known to be a key regulator of inducible hematopoiesis and inflammation. IL-3 is known to stimulate the production and function of basophils and mast cells in the context of allergic inflammation in vivo, and is commonly used as a stimulus to culture these cell types in vitro. In addition, IL-3 signaling has emerged as a potential therapeutic target for the treatment of allergic diseases, including asthma, and hematopoietic malignancies.
Description | Interleukin-3 Human Recombinant produced in E.Coli is a single, non-glycosylated polypeptide chain containing 133 amino acids and having a molecular mass of 15000 Dalton. The IL-3 is purified by proprietary chromatographic techniques. |
Protein Content | Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm using the absorbency value of 0.84 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard solution of IL-3 as a Reference Standard. |
Synonyms | MCGF (Mast cell growth factor), Multi-CSF, HCGF, P-cell stimulation factor, IL-3, MGC79398, MGC79399. |
Source | Escherichia Coli. |
Amino Acid Sequence | The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Pro-Met-Thr-Gln. |
Purity | >96% as determined by SDS-PAGE and HPLC |
Formulation | Sterile Filtered White lyophilized (freeze-dried) powder. Lyophilized from a concentrated (1mg/ml) solution in water containing 0.3 mg/ml of NaHCO3. |
Stability | The lyophilized form of the protein is stable at room temperature for 3 weeks and at -20°C for at least 2 years from the date of receipt. Upon reconstitution, the protein should be stored at 4°C for no more than 7 days. The protein is stable at -20°C for six months. For long term storage, please store at -80°C and the addition of a carrier protein (0.1% HSA or BSA) is recommended. Please prevent freeze-thaw cycles. |
Biological Activity | The ED50 as determined by the dose-dependant stimulation of TF-1 cells is < 0.1 ng/ml, corresponding to a Specific Activity of 10,000,000 IU/mg. |
Solubility | It is recommended to reconstitute the lyophilized Interleukin-3 in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions. |
Usage | LABORATORY RESEARCH USE ONLY. Not intended for diagnostic or therapeutic purposes. |
